ABSTRACT

The objective of this study was to evaluate the usefulness of the China-PAR equations in predicting the 10-year risk of cardiovascular disease (CVD) in the Inner Mongolians population. A population-based, prospective cohort of 2,589 Mongolians were followed up from 2003 to 2012. Participants were categorized into 4 subgroups according to their 10-year CVD risks calculated using the China-PAR equations: < 5%, 5%-9.9%, 10%-19.9%, and ⪖ 20%. The China-PAR equations discriminated well with good C statistics (range, 0.76-0.86). The adjusted hazard ratios for CVD showed an increasing trend among the 4 subgroups (P for trend < 0.01). However, the China-PAR equations underestimated the 10-year CVD risk in Mongolians, and the calibration was unsatisfactory (Hosmer-Lemeshow χ2 = 19.98, P < 0.01 for men, χ2 = 46.58, P < 0.001 for women). The performance of the China-PAR equations warrants further validation in other ethnic groups in China.

ABSTRACT

OBJECTIVE: To investigate the effect of transgelin 2 on paclitaxel resistance, migration and invasion in MCF-7/PTX cells. METHODS: The morphology of MCF-7/S and MCF-7/PTX cells was observed under light microscope. Cell viability was determined using MTT method. The protein and mRNA expressions of transgelin 2, EMT markers including E-cadherin, N-cadherin and Vi-mentin in breast cancer cells were detected by Western blot assay and real-time PCR assay. Wound healing scratch assay and transwell invasion assay were performed to analyze migratory and invasive capability of cells, respectively. Transgelin 2 was reduced in MCF-7/PTX cells by transfecting with TAGLN2 small interference RNA (siRNA). RESULTS: EMT process existed in MCF-7/PTX cells and these cells achieved a high degree of resistance to paclitaxel, and possessed strong ability of migration and invasion. TAGLN2 siRNA treatment sensitized the MCF-7/PTX cells to paclitaxel, and inhibited migration and invasion. CONCLUSION: Overexpression of transgelin 2 could not only induce the paclitaxel resistance, but also inhibit migration and invasion in MCF-7/PTX cells, suggesting that transgelin 2 may serve as a novel biomarker and therapeutic target for breast cancer.

ABSTRACT

<p><b>BACKGROUND</b>In prokaryotic organisms, the mechanism responsible for the accurate partition of newly replicated chromosomes into daughter cells is incompletely understood. Segregation of the replication terminus of the circular prokaryotic chromosome poses special problems that have not previously been addressed. The aim of this study was to investigate the roles of several protein components (MreB, MreC, and MreD) of the prokaryotic cytoskeleton for the faithful transmission of the chromosomal terminus into daughter cells.</p><p><b>METHODS</b>Strain LQ1 (mreB::cat), LQ2 (mreC::cat), and LQ3 (mreD::cat) were constructed using the Red recombination system. LQ11/pLAU53, LQ12/pLAU53, LQ13/pLAU53, LQ14/pLAU53, and LQ15/pLAU53 strains were generated by P1transduction of (tetO) 240 -Gm and (lacO) 240 -Km cassettes from strains IL2 and IL29. Fluorescence microscopy was performed to observe localization pattern of fluorescently-labeled origin and terminus foci in wild-type and mutant cells. SOS induction was monitored as gfp fluorescence from PsulA-gfp in log phase cells grown in Luria-Bertani medium at 37°C by measurement of emission at 525 nm with excitation at 470 nm in a microplate fluorescence reader.</p><p><b>RESULTS</b>Mutational deletion of the mreB, mreC, or mreD genes was associated with selective loss of the terminus region in approximately 40% of the cells within growing cultures. This was accompanied by significant induction of the SOS DNA damage response, suggesting that deletion of terminus sequences may have occurred by chromosomal cleavage, presumably caused by ingrowth of the division septum prior to segregation of the replicated terminal.</p><p><b>CONCLUSIONS</b>These results imply a role for the MreBCD cytoskeleton in the resolution of the final products of terminus replication and/or in the specific movement of newly replicated termini away from midcell prior to completion of septal ingrowth. This would identify a previously unrecognized stage in the overall process of chromosome segregation.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the saponin in Shengmai injection.</p><p><b>METHOD</b>On the basic of studing the chemical constituents of red ginseng and Shengmai injection, 20 compositions had been identifided by LC-MS/MS.</p><p><b>RESULT</b>Twenty identifided compositions were the common components of Shengmai injection and red ginseng extracts.</p><p><b>CONCLUSION</b>The analytical method for saponins in Shengmai injection was established which could be used as the basis for further study and quality control.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To set up a new analysis method of the traditional Chinese medicine Scorpio.</p><p><b>METHODS</b>Ten Scorpio samples were obtained from Hubei, Shaanxi, and Shandong provinces and analyzed with X-ray diffraction Fourier pattern technique to obtain the geometric topology and characteristic marked peak of Scorpio.</p><p><b>RESULTS</b>The geometric topologies of 9 samples were similar, excepting Sample 7#. Totally 11 characteristic marked peaks were observed among these 9 samples.</p><p><b>CONCLUSION</b>X-ray diffraction Fourier pattern is a useful tool for the identification and quality control of the Scorpio.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the effect of Jingui Shenqi pill (JGSQP) with various concentrations at different time points on pituitary adrencorticotropic hormone (ACTH) gene expression level in Shen-Yang deficiency rats.</p><p><b>METHODS</b>The Shen-Yang deficiency rats were randomly divided into the model control group and the high, medium and low dosage of JGSQP groups. Reverse transcriptase polymerase chain reaction was used to observe the effect of JGSQP on the ACTH mRNA of pituitary tissue in rats treated at different time points (10 d, 20 d, 30 d).</p><p><b>RESULTS</b>As compared with that in the model group, the ACTH gene expression level was significantly higher in the high dose JGSQP group (P < 0.05), and the increment in the medium dosage group was significantly higher in comparing with that in the high and low dosage groups (P < 0.05 or P < 0.01).</p><p><b>CONCLUSION</b>Through up-regulation on ACTH gene expression is possibly one of the mechanisms of JGSQP in treating Shen-Yang deficiency.</p>
